Industry Challenge: Unique Specificity

Biomedical researchers employ a variety of methods for evaluating a healthy or naïve cell’s response to an immunogen. MHC-peptide complexes are displayed on the surface of disease infected, cancerous or vaccinated cells as the immune system's alert mechanism to activate a defensive response (Figure 6). Monitoring the downstream signals generated (i.e., cytotoxic T-lymphocytes or soluble signaling molecules, such as cytokines, chemokines, and circulating antibodies) has been the standard up to now; however, this approach only indirectly considers the immune response and does not offer solutions for comparative results. Researchers require new and more specific tools to characterize antigen processing and presentation.

The Receptor Logic Solution

TCRm antibodies exhibit affinity and specificity that exceeds those of the T-cell receptor (Figure 7). These antibodies are stable, predictable and soluble reagents, making them a powerful alternative to cell based assays. TCRms may be used widely in immunological research, including vaccine development (Figure 5: Potency Assay Services).

Receptor Logic's TCRm technology enables direct interrogation of the antigen processed by an antigen presenting cell by revealing the elements, i.e. MHC-peptide complexes, that create the immune response before downstream indicators, such as cell specific responses, reach detectable levels. Many researchers use cell based assays to explore these mechanisms, but these often provide poorly quantitative or comparable results. Receptor Logic’s TCRm antibodies offer significant advantages and capabilities over cell based assays as demonstrated in the Table below.

Advantages / Capabilities of TCRm Compared with Traditional Cell Based Assays

By employing Receptor Logic's TCRm antibodies, researchers can now study and measure the immunological response, while improving the information obtained from existing cell based assays.

Receptor Logic’s TCRm products recognize many peptide antigens used in discovery and product development research. TCRms can immediately integrated manually into and provide a quantitative baseline for a number of established assays, including:

  • Immunocytochemical staining
  • Flow cytometry
  • ELISA
  • Antigen priming
  • ELISpot
  • Cytotoxic T lymphocyte assays
